The recommended dose of tucatinib is 300 mg taken orally twice daily. As the tablets are available in 150 mg strength, this corresponds to two 150 mg tablets per dose. If dose reduction is required due to side effects, your doctor will prescribe 50 mg strength tablets. You may take the tablets with or without food, but they must be swallowed whole, without chewing or crushing, to ensure proper release of the active ingredient in the body. To maintain stable blood concentrations, doses should be taken approximately 12 hours apart, preferably at the same time each day, for example at 8 a.m. and 8 p.m.
If you forget to take a dose, do not take a double dose to make up for the missed one. Simply take your next scheduled dose at the usual time. If you accidentally take an overdose of tucatinib, contact your doctor or pharmacist immediately and show them the medication package if possible. If vomiting occurs after taking a dose, do not take another dose, as the drug may have been partially absorbed. The correct approach is to wait and take your next scheduled dose as planned. Each 300 mg dose of tucatinib (i.e., a single intake) contains 55.3 mg of sodium (equivalent to 2.75% of the daily recommended sodium intake) and 60.6 mg of potassium. If you have renal impairment or are following a low-sodium or low-potassium diet recommended by your doctor, you must take this additional intake into account and inform your doctor. Regarding driving and operating machinery, tucatinib is not expected to directly affect these activities. However, you should make a judgment based on your own physical response. If you experience fatigue, dizziness, or any other discomfort that may affect your concentration while taking the medication, you should avoid driving or engaging in tasks that require a high level of attention.
