Drug Interactions and Contraindications of Tivozanib

When combined with certain drugs, the efficacy of Tivozanib may be reduced or the risk of side effects may increase. Medications that require special notification to the doctor include: Dexamethasone (for anti-inflammatory and immunomodulatory purposes), Rosuvastatin (a cholesterol-lowering drug), Phenobarbital, Phenytoin, Carbamazepine (antiepileptic drugs), Nafcillin, Rifampin, Rifabutin, Rifapentine (antibiotics). Among these, St. John's Wort (a herbal medicine commonly used to treat mild to moderate depression) is strictly prohibited from being used together with Tivozanib, as this herb significantly reduces the blood concentration of Tivozanib, leading to treatment failure. Additionally, if you are taking any hormonal contraceptives (oral contraceptives, implants, or patches), you must use an additional barrier method of contraception (such as condoms) throughout the Tivozanib treatment period and for one month after stopping the drug, because Tivozanib may affect the efficacy of hormonal contraceptives.

Indications and Mechanism of Action of Tivozanib

Tivozanib is used to treat advanced kidney cancer in adults. It is specifically indicated for patients who have not previously received treatments such as interferon-α or interleukin-2, or for whom these treatments have failed to adequately control the disease. Its mechanism of action involves blocking a protein called vascular endothelial growth factor (VEGF), thereby inhibiting the formation of new tumor blood vessels, reducing the blood supply to the cancer, and ultimately slowing the growth and spread of cancer cells.

Important Warnings and Contraindications for Tivozanib

Before taking Tivozanib, the following contraindications must be noted: Do not use if you are allergic to Tivozanib or any of its excipients; it is also prohibited to use if you are taking St. John's Wort (a herb used to treat depression and anxiety). In addition, patients should inform their doctor of their complete medical history, including hypertension, blood clots, bleeding, liver disease, thyroid disease, etc. Pregnant women must not take this drug, and reliable contraceptive measures should be taken during treatment and for one month after stopping the drug. Breastfeeding women should also avoid breastfeeding.
