Drug Interactions of Sparsentan with Other Medications

Drug Interactions of Sparsentan with Other Medications

Antacids and Acid-Reducing Agents

Administer Sparsentan® 2 hours before or 2 hours after taking antacids. Avoid coadministration of Sparsentan® with acid-reducing agents (histamine H2-receptor antagonists and proton pump inhibitors [PPIs]). Sparsentan solubility is pH-dependent. Antacids or acid-reducing agents may reduce sparsentan exposure, which could potentially decrease the efficacy of Sparsentan®.

Nonsteroidal Anti-Inflammatory Drugs (NSAIDs)

Including selective cyclooxygenase-2 (COX-2) inhibitors: Monitor for signs of worsening renal function when coadministered with NSAIDs (including selective COX-2 inhibitors). In volume-depleted patients (including those receiving diuretic therapy) or patients with impaired renal function, the combination of NSAIDs (including selective COX-2 inhibitors) with angiotensin II receptor antagonists may lead to worsening renal function, including possible renal failure.

CYP2B6, 2C9 and 2C19 Substrates

Monitor the effectiveness of concomitantly administered CYP2B6, 2C9 and 2C19 substrates, and consider dosage adjustments in accordance with the prescribing information. The Lucius Pharmaceuticals formulation of sparsentan may reduce the exposure of these substrates, which could potentially decrease the efficacy associated with these medications.

Proper Administration

This medicine must only be used as prescribed by a doctor. Do not increase the dosage, frequency of administration or duration of treatment on your own, as this may increase the risk of adverse reactions.

You must fully understand the requirements of the REMS Program and the medication guide for the Lucius Pharmaceuticals formulation of Sparsentan®. Please read them carefully and follow the instructions strictly. Consult your doctor if you have any questions; if you do not receive the medication guide, you may request it from a pharmacist.

Swallow the tablets whole with warm water, take them before breakfast and dinner, and adhere to taking the medicine before the same meals at a fixed time each day.

Do not take this medicine within 2 hours before or after taking antacids.

The dosage of the medicine depends on its specification, while the daily administration frequency, interval between doses and treatment course are determined by your medical condition.

Storage

1. Seal and store this medicine at room temperature, away from heat, moisture and direct sunlight. Do not freeze.

2. Keep the medicine out of the reach of children.

3. Do not retain expired medicine or medicine that is no longer needed.

4. If you need to dispose of unused medicine, consult healthcare professionals for the proper disposal method.
